The effect of vitamin D3 administration and its combination with physical activity on oxidative stress markers in old rats

Abstract

Background. The natural phenomenon of aging is accompanied by several biological changes, including increased oxidative stress (OS). The aim of the study was to observe how vitamin D3, physical activity and their combination affect oxidative stress in the aging organism.

Groups and methods. We included 48 male Wistar rats in the experiment. The control group consisted of adult rats aged 6-8 months (AR), which received a placebo daily for 7 weeks. The old rats aged 25-27 months were divided into 7 groups: C3/C7 – placebo daily for 3/7 weeks; D1 – vitamin D3 at a single dose of 800 IU/kg of body weight (BW)/month; D2 – vitamin D3 at a daily dose of 14 IU/kg of BW for 3 weeks; Ex – placebo daily for 3 weeks + 4 weeks daily placebo and exercise; D1Ex – vitamin D3 at a single dose of 800 IU/kg of BW/month + vitamin D3 at the same single dose and 4 weeks of exercise; D2Ex – vitamin D3 at a daily dose of 14 IU/kg of BW for 3 weeks + 4 weeks daily vitamin D3 and exercise. We determined biochemical parameters and OS markers in the plasma, and activities of antioxidant enzymes in erythrocyte hemolysate.

Results. The administration of a single high dose of vitamin D3 resulted in an increase in the testosterone levels, total antioxidant capacity TEAC (Trolox equivalent antioxidant capacity) and a decrease of 8-isoprostane levels. Advanced oxidation protein products (AOPP) levels and TEAC increased after a single high dose of vitamin D3 combined with physical activity. Daily administration of lower dose of vitamin D3 decreased 8-isoprostane levels and, in combination with physical activity, it decreased total cholesterol levels, glutathione peroxidase activity and increased TEAC. Physical activity alone decreased glucose levels and increased TEAC.

Conclusion. Physical activity, vitamin D3 and their combination increased the antioxidant capacity and can thus contribute to the reduction of oxidative stress in the aging organism (Tab. 3, Ref. 45). Text in PDF www.lekarskyobzor.sk.
